The fixation of many to define rules to define the boundaries of the nebulously-described “drug like chemical space” has been overly influenced by Lipinski’s seminal “Rule of 5.” This lecture will illustrate why it is time to move on from and beyond the confines of these “approaches to estimate solubility and permeability” with insights on contemporary principles of optimum, efficiency and physicochemical properties. Important compromises in drug discovery are often made to ensure optimal permeation of membranes, an area of some controversy; theories will be reviewed and suggestions made for principles applicable to antimicrobial research in particular.
